Abstract

Elacyt is a cytotoxic agent being tested as a treatment for haematologic cancers and solid tumors. To enhance cellular drug uptake, lipophilic analogs (5'-elaidic acid esters) of cytarabine and gemcitabine were synthesized. Elacyt (5'-. O-(trans-9'-octadecenoyl)-1-β-d-arabinofuranosyl cytosine) has a longer cellular retention and a different effect on DNA and RNA synthesis than cytarabine. Elacyt has distinct antitumor activity in various solid tumour xenografts in which cytarabine is inactive. In phase I trials, elacyt displayed a favorable safety profile. Activity has been demonstrated in melanoma, NSCLC, and lung cancer refractory to gemcitabine treatment (Breistøl, 1999) ... © 2008
