Abstract
Tipifarnib, an orally administered non-peptidomimetic farnesyl transferase inhibitor, has shown promising clinical activity. Farnesyl transferase inhibitors are a novel class of anticancer agents that competitively inhibit farnesyltransferase, an enzyme that is essential for farnesylation. Farnesylation leads to oncogene Ras activation, and to the development of neoplasms. The mechanism of action of this class of anticancer drugs appears complex, involving proteins in addition to Ras. Preliminary results from clinical trials demonstrate enzyme target inhibition, low toxicity and promising clinical activity. .... . © 2009
